Preparation
Virus extracted from infected cells is concentrated by ultracentrifugation and then partially purified by centrifugation through sucrose. The antigen is resuspended and homogenised. The resulting preparation consists of a high concentration of virus and viral components as well as some cellular material suspended in 0.1 M glycine buffer pH 9.5.
Inactivation
Beta-propiolactone treatment. Inactivity is confirmed by attempted growth under original culture conditions. Since no procedure can guarantee absolute sterility, the reagent should be handled with appropriate safety precautions.
